高效液相色谱法测定饮料中的咖啡因含量

摘要:咖啡因是咖啡饮料、茶饮料和可乐等饮料中的主要成分,也是中枢神经系统的兴奋药。本文建立了高效液相色谱测定饮料中咖啡因含量的方法,色谱柱为反相ODS C18 柱,甲醇—水溶液为流动相进行洗脱。结果表明,在柱温35℃,甲醇∶水为1∶1,流速1.0mL/min,检测波长272nm条件下,咖啡因的浓度在0.1~70.0mg/L范围内与峰面积成良好的线性关系,回收率为79.79%~101.56%,精密度RSD 在0.11%~1.38%。该方法可快速、准确地测出各种市售饮料中咖啡因的含量。关键词:高效液相色谱;饮料;咖啡因

Determination of Caffeine Concentration in Beverage by High Performance Liquid Chromatography

Abstract: Caffeine is a major ingredient of daily beverages including coffee,tea,cola,et al,and is the stimulant of centralof nervous system as well as one of most widely used drugs in the world. The method was developed to determine ofcaffeine concentration in beverages using high performance liquid chromatography (HPLC)with ultraviolet detector.Reversed phase liquid chromatographic separation was performed on ODS C18 column,The mobile phase was amixture of methanol and deionized water. Results showed that the optimum conditions were methanol-water(1∶1,v/v)and the flow rate 1.0ml/min,the column temperature 35℃ and detecting wavelength 272nm. Under the optimizedconditions,the linearity of caffeine was very good in the considered concentration ranges of 0.1~70mg/L(r2=1). Therate of recovery for spiked standards (n=7)ranged from 79.79% to 101.56%,showed good accuracy of proposedanalytical method. Average relative standard deviations(RSDs)were 0.11~1.38%. This method is simple and accuratein determination of caffeine concentration in beverages and satisfactory results can be obtained.Key words: high performance liquid chromatography;beverage;caffeine
