HPLC 法测定饮料中的果糖、葡萄糖、麦芽糖、乳糖含量

摘要:建立了高效液相色谱-蒸发光散射法测定饮料中果糖、葡萄糖、麦芽糖及乳糖含量的方法。用水作为提取剂,以乙腈-水(体积比70∶30)为流动相,采用Ultimate® XB-NH2柱(250mm×4.6mm,5μm)进行分离,蒸发光散射检测器检测。结果表明:4种糖在2.0~10.0mg/mL均有良好的线性关系,相关系数(r)为0.9999;方法检出限为0.33g/kg~0.82g/kg;3 个不同水平(2.0、4.0、8.0g/kg)的加标平均回收率为96.00%~102.50%,RSD值为1.14%~2.04%。该方法前处理简单、灵敏度高、重现性佳、检测速度快,适用于饮料中4 种糖的同时检测。

Determination of Fructose,Glucose,Maltose and Lactose in Beverages by High PerformanceLiquid Chromatography

Abstract: To establish a method for determination of fructose,glucose,maltose and lactose in beverages by highperformance liquid chromatography- evaporative light scattering(HPLC- ELSD). The samples were extracted withwater. The separation of targeted compounds was performed on a Ultimate® XB-NH2 column(250mm×4.6mm,5μm)using acetonitrile-water(70∶30,v/v)as mobile phase,detected by evaporation light scattering detector(ELSD). Theresults showed that the 4 kinds of sugars had good linearity in the concentration range of 2.0~10.0mg/mL,and thecorrelation coefficients were 0.9999;the limit of detection was 0.33g/kg~0.82g/kg;and the standard recovery rate(standard additive concentration is 2.0,4.0,8.0g/kg) was between 96.00% ~102.50% ,and relative standarddeviation(RSD)was from 1.14% to 2.04%. This method has the advantages of simple pretreatment,high sensitivity,good reproducibility and fast detection speed,which is suitable for quantitative detection of fructose,glucose,maltose and lactose in beverages.
