气相色谱法测定乳与乳制品以及植物蛋白 饮料中胆固醇的含量

摘要:建立了乳与乳制品以及植物蛋白饮料中胆固醇的气相色谱仪测定方法。样品经无水乙醇和氢氧化钾溶液皂化,石油醚和无水乙醚混合提取,提取液浓缩至干,无水乙醇溶解定容,采用氢火焰离子化检测器检测,外标法定量。在DB-5 弹性石英毛细管柱程序升温分离胆固醇,流速为1mL/min;进样口温度:280℃;检测器温度:290℃;进样模式:分流模式,分流比1∶30;氢气流速:40mL/min;空气流速:400mL/min。每100g 乳与乳制品以及植物蛋白饮料中胆固醇的添加量为1mg~40mg时,加标回收率为90.5%~94.5%,本方法的方法检出限:0.3mg/100g,定量限:1mg/100g。该方法对皂化温度要求比较高,具有结果准确、处理方法快速简便、重复性好等优点,可用于乳与乳制品以及植物蛋白饮料中胆固醇的检测。

The Content of Cholesterol in Milk and Dairy Products and Vegetable Protein Beverage wasDetermined by Gas Chromatography

Abstract: A method for the determination of cholesterol in milk and dairy products and vegetable protein beverage bygas chromatograph was established.The sample is saponified by anhydrous ethanol and potassium hydroxide solution,and extracted by mixing petroleum ether and anhydrous ether. The extract is concentrated to dry and dissolved inanhydrous ethanol,and then the sample was detected by hydrogen flame ionization detector and quantified by externalstandard method. Cholesterol was separated by temperature rising in DB-5 elastic quartz capillary column. The carriergas was high purity nitrogen and the flow rate was 1mL/min.Inlet temperature: 280℃;Detector temperature:290℃;Injection mode:split mode,split ratio 1∶30;Hydrogen flow rate:40mL/min;Air flow rate:400mL/min. When theadded amount of cholesterol in milk,dairy products and vegetable protein drinks is 1mg~40mg per 100g,the standardrecovery rate is 90.5%~94.5%. Detection limit of this method is 0.3mg/100g,and the quantitative limit is 1mg/100g.This method requires high saponification temperature,has the advantages of accurate results,quick and simpletreatment,good repeatability,and can be used for the detection of cholesterol in milk and dairy products and vegetableprotein beverage.
