摘要:建立了基于分光光度法测定原料中透明质酸钠的方法。用硼砂作催化剂对透明质酸钠用硫酸进行酸解,在100℃水浴震荡20min,通过酸解可以把葡萄糖醛酸分解出来,再加入咔唑在100℃水浴震荡20min 进行衍生,衍生液在分光光度计上分析。当透明质酸钠的添加量在4.5g/100g~210.0g/100g 时,加标回收率为95.3%~104.5%,相对标准偏差小于3.0%,定量检出限为10.0g/100g。该方法具有快速、准确、稳定性好、灵敏度高等优点,可用于原料中透明质酸钠含量的检测。

Determination of Sodium Hyaluronate in Raw Materials by Spectrophotometry

Abstract: A method for the determination of the content of sodium hyaluronate in raw materials byspectrophotometry was developed. Acidulated sodium hyaluronate with sulfuric acid using borax as catulyst andshaked in water bath at 100℃ for 20min,the glucuronic acid can be broken down by acidolysis. The 0.1% carbazoleethanol solution was added and shaken in water bath at 100℃ for 20mins for devivatization. The derivative solutionwas analyzed on a spectrophotometer.Added sodium hyaluronate content was in the range of 4.5̆5̆~210.0g/100g,theaverage recoveries were between 95.3% and 104.5%,with RSDs(n=6)less than 3.0%. The limit of quantitation was10.0g/100g. The results indicated that this method is accurate,simple and reproducible,and thus suitable for theanalysis of the content of sodium hyaluronate in raw materials.
