摘要:为研究玉米低聚肽对肝纤维化的影响,以昆明小鼠为研究对象,灌胃CCl4 建立肝纤维化模型,灌胃不同剂量的玉米低聚肽溶液给予治疗,同时给予1mg/kg 剂量的秋水仙碱作为阳性对照。通过苏木素-伊红(hematoxylineosin,HE)染色和Masson 三色染色评估肝纤维化小鼠的肝损伤和胶原沉积;通过酶联免疫吸附测定法(enzymelinked immunosorbent assay,ELISA)、反转录聚合酶链反应(reverse transcription PCR,RT-PCR)和蛋白免疫印迹法(western blotting,WB)研究玉米低聚肽对肝纤维化的改善机制。HE 染色和Masson 三色染色结果表明玉米低聚肽能够使受损的肝组织结构恢复正常,且减少了肝组织胶原沉积。此外,ELISA、RT-PCR 和WB 结果表明玉米低聚肽通过降低促炎因子水平和减少细胞外基质的产生从而改善肝纤维化。上述结果表明玉米低聚肽具有护肝的效果,且基于玉米低聚肽的护肝效果进一步研究一种以玉米低聚肽为核心原料的有益于肝脏的饮料配方,为其作为新型益肝饮料的开发提供了理论支撑。

Study on the Mechanism of Corn Oligopeptides Improving Liver Fibrosis and Its Application in LiverProtection Drink

Abstract: To study the effect of corn oligopeptides(COPs)on liver fibrosis,Kunming(KM)mice were given CCl4intragastric administration to establish liver fibrosis model. COPs solution at different doses was given intragastricadministration,and colchicine at 1mg/kg was given as positive control. Hematoxylin-eosin(HE)staining and Massontrichrome staining were used to evaluate liver injury and collagen deposition in liver fibrosis mice. The improvementmechanism of COPs on liver fibrosis was investigated by enzyme linked immunosorbent assay (ELISA),reversetranscription PCR(RT-PCR)and western blotting(WB). COPs could restore the damaged liver tissue structure tonormal and reduce collagen deposition in liver tissue as suggested by HE staining and Masson trichrome staining. Inaddition,the results of ELISA,RT-PCR and WB showed that COPs improved liver fibrosis by reducing proinflammatorycytokine levels and extracellular matrix production. The above results indicated that COPs have the effect of protecting liver. And based on the liver protecting effect of COPs,a liver protection drink formula with COPs as thecore raw material was further studied,which provides theoretical support for its development as a new type of liverprotecting drink.
