高效液相-质谱法测定饮料中5 种糖和3 种糖醇

摘要:目的:建立了一种高效液相色谱-质谱法检测饮料中阿拉伯糖、果糖、葡萄糖、蔗糖、麦芽糖、赤藓糖醇、山梨糖醇和甘露糖醇含量的方法。方法:样品经涡旋混匀,超声提取和稀释后,经高效液相色谱串联质谱分析。流动相为A 相(90 %乙腈+5 %水+5 %异丙醇),B 相(80 %乙腈+20 %水),梯度洗脱,流速为0.85mL/min,柱温为65 ℃,样品进样量为1μL,以保留时间选择离子定性,以外标法定量。结果:阿拉伯糖、果糖、葡萄糖、蔗糖、麦芽糖、赤藓糖醇、山梨糖醇和甘露糖醇在2μg/mL ~100μg/mL 质量浓度范围之间具有良好的线性关系(R2>0.99);饮料的加标回收率在94.40%~111.94%;方法精密度相对标准偏差RSD%(n=6)在0.68%~4.62%。果汁的加标回收率在90.51%~109.94%;方法精密度相对标准偏差RSD%(n=6)在0.71%~4.82%。结论:本方法可应用于饮料中阿拉伯糖、果糖、葡萄糖、蔗糖、麦芽糖、赤藓糖醇、山梨糖醇和甘露糖醇的测定。

Determination of 5 Sugars and 3 Sugar Alcohols by High Performance Liquid Chromatography-MassSpectrometry Method

Abstract: Objective:To develop a high performance liquid chromatography-mass spectrometry(HPLC-QDa)methodfor the determination of arabinose,fructose,glucose,sucrose,maltose,erythritol,sorbitol and mannitol in drink andjuice. Methods:Sample was extracted by vortex oscillated,ultrasonic extracted and diluted with extraction solution,then analyzed by high performance liquid chromatography-mass spectrometry(HPLC-QDa). The mobile phase A was90% acetonitrile-5% water-5% isopropanol(V/V/V). The mobile phase B was 80 % acetonitrile -20% water(V/V).The gradient elution was carried at a flow rate of 0.85mL/min and the column temperature at 65℃,and the sampleinjection volume was 1μL. Sugar and sugar alcohol were qualitatively determined by retention time and selected ions,and were quantified by external standard method. Results:The linear relationship of arabinose,fructose,glucose,sucrose,maltose,erythritol,sorbitol and mannitol was good(R2>0.99)in the concentration range of 2-100μg/mL. Thespiked recoveries of drink are within the range of 94.40%~111.94%. The relative standard deviation(RSD) ofmethod precision was 0.68%~4.62%(n=6). And the spiked recoveries of juice are within the range of 90.51%~109.94% . The relative standard deviation(RSD) of method precision is 0.71% ~4.82%(n=6). Conclusion:Thismethod is applicable for the determination of arabinose,fructose,glucose,sucrose,maltose,erythritol,sorbitol andmannitol in drink.
